Alk Cancer
Alk positive cancer is a type of cancer with no known cause and no known cure once it has spread to other parts of the body. Younger patients usually 55 and under who have never smoked are most likely to be diagnosed as being alk.
Alk positive cancer occurs in humans mostly as lung cancer but it can also originate in many other parts of the body including brain and breast.
Alk cancer. Anaplastic lymphoma kinase alk was originally discovered in 1994 in anaplastic large cell lymphoma alcl cells. Alcl is caused by a 2 5 p23 q35 chromosomal translocation that generates the fusion protein npm alk in which the kinase domain of alk is fused to the amino terminal part of the nucleophosmin npm protein. It is thought to occur in about 100 000 people worldwide every year.
When an elevated alp level is first detected via routine blood testing it usually prompts a more thorough investigation to determine potential causes. It is not thought to be a disease that is inherited or can be passed on to others in any way. They fall under the category of tyrosine kinase inhibitors which work by inhibiting proteins involved in the abnormal growth of tumour cells.
More than 19 different alk fusion partners have been discovered in nsclc including eml4 kif5b klc1 and tpr. Alk inhibitors are anti cancer drugs that act on tumours with variations of anaplastic lymphoma kinase alk such as an eml4 alk translocation. Recent research retrospectively analyzed clinical outcomes for 110 patients with alk positive stage iv nsclc who were treated at the university of colorado cancer center in aurora between 2009 and.
Alk positive anaplastic lymphoma kinase positive or alk lung cancer occurs in 1 out of 25 non small cell lung cancer patients nsclc the most common type of lung cancer. The mutation is a gene rearrangement an abnormal fusion of alk and another gene echinoderm microtubule associated protein like 4 eml4. However it occurs in approximately 30 of lung cancer patients diagnosed under age 40.
It is caused by a mutation of the alk gene. Lorlatinib a third generation inhibitor of anaplastic lymphoma kinase alk has antitumor activity in previously treated patients with alk positive non small cell lung cancer nsclc. Alk positive lung cancer occurs in approximately 5 of all lung cancer patients.
Alk positive lung cancer is a type of non small cell lung cancer nsclc in which the cancer cells have a mutation in the anaplastic lymphoma kinase alk gene. Alk first emerged in the field of oncology in 1994 when it was identified to fuse to npm1 in anaplastic large cell lymphoma. Conversely cancer victims who are responding to treatment will typically see a fall in their alp levels which is a positive prognostic sign.
The efficacy of lorlatinib as compared with that of crizotinib as first line treatment for advanced alk positive nsclc is unclear. Since then alk has been associated with other types of cancers including non small cell lung cancer nsclc.
